Abstract

The glycolytic particle, a conglomerate of glycolytic enzymes was first speculated upon over half a century ago with supporting evidence accumulating quite rapidly thereafter. Early studies used immunohistochemistry to locate enzymes in the cell and to pull down binding partners with affinity to immobilized enzymes and/or proteins. Whilst early studies largely looked at binary interactions, advances in microscopy and mass spectrometry of proteins and metabolites have since enabled pathway level studies. As one of the canonical pathways harbouring a metabolon, glycolysis has been subject to a particularly wide range of methods spanning most of those described within this book. Here we summarize the application of these methods and describe how they have illuminated our understanding of the regulation of glycolysis, utilizing this pathway as an example for the study of the metabolon. We conclude by stating that while there is categorical evidence supporting the presence of glycolytic metabolons across kingdoms, we now need to better address the questions of their physiological relevance and their role in the molecular hierarchy underlying the regulation of this cardinal pathway.
