Abstract

An overview on the various optical-based methods is given which shows the characterization of the assembly and turnover of macromolecular complexes that are present at neuronal contacts. The biological paradigm and some relevant molecular components has been explained while microscopy techniques have been described to show a dynamic visualization of specific elements at such contact sites. A focus is given specifically on the following: optical tweezers, FRAP, single-molecule and particle tracking, FCCS, and FRET/FLIM. Mathematical analysis were done on each case to present the physicochemical parameters that define protein-protein interactions.
