This project describes the proteomic profiling of human epilepic brain tissue, alongside control samples. Microdissected hippocampal, neocortical, and white matter tissue was analyzed. The aim was to identify protein alterations associated for different epileptic conditions. Raw mass spectrometry data, along with search engine output, are included in this partial submission. Sample Processing Protocol Fresh-frozen brain tissue was homogenized in lysis buffer and beads. Protein extraction was done by adding lysis and digestion buffer with trypsin. Peptides were labeled with TMT-16-plex reagents for quantification. LC-MS/MS analysis was done by injecting peptides into Orbitrap Eclipse mass spectrometer and full-MS spectra were collected. Data Processing Protocol Raw MS files were processed with Proteome Discoverer 2.4 (Thermo Fisher Scientific). Spectra were matched against Homo Sapiens database obtained from UniProt. All results were filtered to a 1% FDR. Search engine output files and raw mass spectrometry files included.
