Abstract

A variety of operating modes and process configurations exist for designing efficient chromatographic separations. Decisions to bemade concern a pulse-wise or continuous feed introduction, operation in a single- or multi-column mode, elution under isocratic or gradient conditions, co-, cross-, or counter-current flow of mobile and stationary phases, and withdrawal of two or a multitude of fractions. Furthermore, while fine chemicals are of low molecular weight, biomolecules have a molecular weight up to several million and may undergo a denaturation. Correspondingly, the type of the molecules to be separated is constraining process options and may require very different process designs.
